What is a peptide reference standard?
A reference standard is material characterized and assigned for a defined analytical use. It may establish identity, calibrate content, support impurity measurement or monitor method performance. The certificate must say which value is assigned and on what basis. A peptide with an HPLC purity result but no content assignment should not be assumed to provide an exact mass concentration.
Peptide standards are challenging because water, counter-ions, residuals, oxidation and adsorption can change the effective amount. Characterization commonly combines identity, purity and quantitative techniques rather than relying on a single chromatogram.
How do primary and secondary standards differ?
A compendial or well-characterized primary standard can anchor a measurement. A working or secondary standard is more accessible for routine use but must be qualified against the chosen reference under a controlled procedure. Qualification should cover the attribute used in the method and define requalification or expiry.
Traceability is a documented comparison chain, not a marketing adjective. Record standard lot, storage, preparation, assigned value, uncertainty where applicable and each analytical sequence in which it was used.
What is system suitability?
System suitability is a set of pre-run or sequence-level checks showing that the analytical system can perform the required separation or measurement. HPLC criteria may cover retention, replicate injection precision, peak shape and resolution between critical components. LC-MS criteria may include mass accuracy, sensitivity and carryover.
The sample result should not be used to declare the system suitable after the fact. Criteria are selected from method-development and validation knowledge, then applied before or around the sample sequence according to the procedure.
How can laboratories avoid false confidence?
Do not use one vial indefinitely without monitoring degradation or evaporation. Avoid repeatedly thawing and returning the same standard. Compare new working-standard lots with an established lot across the analytical range. Investigate a suitability failure before reporting samples, because reinjection without a documented cause can hide an unstable method.
A standard makes a measurement more comparable only when its assigned value, preparation and lifecycle are controlled. Naming a substance as the standard is not enough.
